Enhancement of granulocyte chemiluminescence with hydroxyl radical scavengers.
نویسنده
چکیده
Addition of hydroxyl radical scavengers (benzoate, ethanol, and mannitol) to granulocyte chemiluminescent reactions significantly enhanced light generation without altering granulocyte oxygen consumption or superoxide production.

Hydroxyl radical scavengers produce similar decreases in the chemiluminescence responses and bactericidal activities of neutrophils.
The addition of hydroxyl radical (.OH) scavengers caused similar decreases in the chemiluminescence responses and killing of Staphylococcus aureus 502A by human neutrophils in vitro.
متن کاملEnhanced chemiluminescence of the luminol-hydrogen peroxide system by colloidal cupric oxide nanoparticles as peroxidase mimic.
As a peroxidase mimic, cupric oxide nanoparticles were found to enhance the chemiluminescence (CL) of luminol-H(2)O(2) system up to 400 folds. The CL spectra and radical scavengers were conducted to investigate the possible CL enhancement mechanism. It was suggested that the enhanced CL could be attributed to the peroxidase-like activity of CuO nanoparticles, which effectively catalyzed the dec...
متن کاملHuman Granulocyte Generation of Hydroxyl
As human leukocytes phagocytize, there is an associated burst of oxygen consumption (1), emission of light (chemiluminescence) (2), stimulation of the hexose monophosphate shunt (1), and the production of a variety of oxygen related toxic agents (3). These events appear to comprise an integral part of the anti-bacterial mechanism (4). Hypothesized mediators of the oxygen-dependent bactericidal ...

Chemiluminescence of luminol in a cell-free system was used to investigate the mechanism of alloxan-dependent free-radical generation. In the presence of alloxan and reduced glutathione (GSH), luminescence was greatly stimulated by FeSO4. Replacing GSH by oxidized glutathione or NAD(P)(H), or replacing FeSO4 by CUS04, ZnSO4 or FeCl3, did not yield chemiluminescence. The chemiluminescence of a m...
متن کاملHuman granulocyte generation of hydroxyl radical
Human granulocytes were capable of oxidizing 2-keto-4 thiomethylbutyric acid to ethylene during phagocytosis or membrane perturbation. The reaction required hydrogen peroxide and superoxide and in addition was inhibited by various hydroxyl radical (OH) scavengers. These observations represent direct evidence for the generation of OH by human granulocytes. Further, inhibition of ethylene generat...
